This study aims to characterise bacterial extracellular vesicles (BEV) released by commensal Bifidobacterium strains and to examine whether these BEV have protective potential to support blood-brain barrier (BBB) integrity and astrocyte activity under inflammatory conditions, which may benefit cognitive function.
The study will assess the impact of BEV on gene expression profiles in human brain endothelial cells of the HBEC5i cell line and evaluate their effects on BBB physiology, with particular attention to the regulation of tight junction protein expression, secretory activity, and the activation of MAPK and PI3K/Akt signalling pathways. In addition, the influence of BEV on the expression of protective factors released by astrocytes will be investigated.
This research aims to improve our understanding of the communication between the microbiota and the nervous system and may, in the future, provide the basis for innovative and targeted therapeutic approaches using specialised next-generation postbiotics to support brain health. Potential applications include improving cognitive function - a major public health concern - and contributing to the prevention/treatment of neurodegenerative disorders
